My top suggestions would be

50's prime time at DHS
Chef Mickey at Contemporary
Yatchmen Steakhouse at the YC (expensive but great steaks)
Boma at AKL (especially if you like to try different tasting foods)
Le Cellier in Canada at Epcot (great steaks here too)

You will have no problem getting suggestions, there are a lot of knowledgeable folks here but a little bit of information might really help with the responses:
1. Are you going alone?
2. Are there children involved?
3. Do you want "character meals"?
4. How much $$ is too much?

My philosophy for dining out (WDW or not) is to experience something new and go somewhere to eat something you wouldn't normally eat. Take into consideration your tastes, budget, party size, character dinners, shows, etc. There are very few place that you can go wrong in WDW. Its just a matter of preference. Me personally, I like (in no particular order) Kona Grill, Ohana, California Grill and Le Cellier. But I like almost all food, so there is no way I could ever go wrong.

I'm going with the idea that if someone told me Disney was closing forever and I had one last trip where I would go.

Epcot: Le Cellier ( the food is amazing as well as the atmosphere)

San Angel Inn ( food is good but nothing beats the atmosphere and sitting and watching the boats from the attraction pass)

Chefs de France ( food and atmosphere)

light breakfast - Kringla or Boulangerie ( Norway or France)

DHS: Sci-Fi and 50's ( just because you have to)

MK: Liberty Tree Tavern ( nothing beats the build up of knowing you are near the Mansion and the two mountains ) food is usually good and I love the colonial theme

Pecos Bill - best burgers in the West - love the fixins' bar - theme in general is great

The only MUST for us is 50's Prime Time Cafe. We've gone every trip (and loved it every time). So, it's kind of a tradition.

Ohana comes close, but it's not a must.

I would say The Brown Derby, but I've read their menu and service has been declining the past couple of years. Too bad, I really like the Cobb Salad.
