No. Smokey is dead. The beginning premise was good, but have to come up with another villain.

Smokey as an embodiment of John Locke or MiB is dead, but they could always find a way to just bring him back for the ride. For instance, when the island was skipping in time, one of the times could have been while Hurley was the leader or someone else goes back into the cave with the light and unleashes a new Smokey. The ride wouldn't necessarily have to entirely follow the shows cannon, though they could easily go with a number of other ideas for the ride.
