Been to Disney many times and never rented a car
looking to rent a car in April for the first time and visit a Uncle in the Villages for the day
whats the best option for me for the day

You could rent from the Car Care Center, but typically I have found the best deal from Avis. They have a desk at the Hilton Lake Buena Vista. The desk closes at 4pm though I think so I usually pick up around noon (get lunch at Disney Springs and walk over) and return the next morning (and take their shuttle to the parks). Some of the other Disney Springs hotels have car rental desks which just shop around a bit.

We rented a car for a couple of days from the DS hotel that is just across the footbridge. I think that could be the same one that @ajrwdwgirl used.

The Disney Car Centre wouldn’t allow us to book for just 2 days, but then we realised we could find one cheaper than the theoretical price Disney we’re charging.

There are several options at the DS resorts.

Just an update: The Wyndham Lake Buena Vista had a Hertz car rental location, but they just recently closed it (with no notice whatsoever to those with reservations there -- I just happened to be poking around online looking at the details for our August 1-day rental that was supposed to be returned at the Wyndham, and noticed that the location was no longer listed on their website, so I called and got the full story... good thing I did, or we'd have been in for a rude awakening when we went to return the car in August!)

Just letting you know that the Hertz at the Disney Springs-area Wyndham is no longer an option, as I imagine it will take awhile for Google and other online resources to reflect the change. There are, as others mentioned, several other nearby car rental places by Disney Springs, however, some within comfortable walking distance.

That is unfortunate for the people with reservations there. We rented from that place a few years ago and it was a convenient place.
