Is there a number which allows you to call these resorts directly to make room requests?

(407) 934-8000, but there’s a lot of debate as to whether any resort really has a direct line (and they probably don’t). It’s most likely a call center that answers “Beach Club” on that number. It doesn’t hurt to try, though. They’ll take your request, but I would make sure to call the main WDW Reservations number as well and make sure they have your requests.
