Tickets and Room booked separate

I am going down next week and have no information how this new fast pass system works. We booked the room with one of our friends vacation club and are staying at the boardwalk villas. However, we ordered our tickets separate and will pick them up at will call. How will the fast pass plus work in this situation and can we even book it ahead of time?

I did the same thing this past trip in November. As soon as I picked up my tickets I took them to the room and added them to my profile on the disney website. Then I selected FP+ for the rides for everyone..everything worked fine.
No you cannot book a FP+ ride until you have the tickets added to your profile.
If you aren't taking a laptop to the resort probably the concierge desk can add them for you, maybe even the will call window will do it.

Did you order your tickets online? While it is true you cannot get your tickets until you arrive at will call. You can input the serial numbers from certificates emailed to you from Disney Vacations and have them logged to your account (see example from my account below). For FP+ purposes, they work just like the actual tickets. So, when you're 60 days or less out from your arrival at your hotel, you can start inputting your FP+ choices.

It depends on the way you ordered the tickets. I have an AP for my son that's "will call" and I couldn't add it to MDE, but my AP voucher they sent me could be.

To the OP:
You mentioned that you booked at a DVC resort through your friend. Do you (and your travel companions) have profiles set up in the My Disney Experience page yet? And if so, have you linked the resort reservation to those profiles? That's a critical step that must be done to a) get your Magic Bands customized, and b) fulfill the first part needed to make FP+ reservations. (The other half being linking your park tickets to the profiles, as others have mentioned)
