From Sanford Airport to Disney - routes? tolls?

If you are driving a hire car the tolls can be charged to your cc on file with the hire company. No need for change, I think know the system is called Sunpas. If you just flew from the UK I would recommend taking the easiest route to WDW which includes the toll roads. It can add up depending on how far you travel, I once spend $20 in one day on tolls but it is much easier to just use the Sunpas lane and charge the toll to the car. Hope this helps.

Weekday or weekend? If it is the weekend I-4 might not be that bad. You'll hit some congestion as you get near the attractions and then again nearing Walt Disney World.

Taking SR 417 from US 19/92 near the airport to Exit 6 International Drive (the exit name is misleading) will cost you $7.25 in cash.

If you don't need a car, we just hired Mears to pick us up in a van and take us to our motel. They usually let you have a half hour store stop if you need anything. It was 140 each way

This. Definitely better not driving if you are staying on property, and switching from UK to US driving is not an activity I'd recommend to anyone on vacation:)
